Evaluation of Kjeldahl Digestion Method

چکیده
The evaluation of the Kjeldahl digestion method was investigated by comparing measured values of total nitrogen, phosphorus and potassium using three salt and catalyst mixture in Standard Kjeldahl digestion method and Salicylic acid Modification method with certified values of plant material as well as comparison was made for determination of total nitrogen from steam distillation method verses the Technicon Auto-analyzer, and phosphorus Ascorbic acid/Molybdate method verses Molybdate/ Metavanadate method on the Technicon Auto-Analyzer. The 1 g salt/catalyst mixture recovered less nitrogen than the 2.5 g in the standard Kjeldahl method due to the lower temperature and incomplete digestion in both plant and soil samples. The 2.5 g catalyst mixture partially recovered nitrate in the standard Kjeldahl method and the salicylic acid modification fail to recover all nitrate in plant material. Use of 2.5 g salt catalyst mixture and selenium appears to promote nitrogen losses in salicylic acid modification method but not in the standard Kjeldahl method of digestion for soil samples. No interference of selenium or copper was observed in Nitrogen and Phosphorus on colorimetric determination. The standard kjeldahl method with 2.5 g of salt/catalyst mixture of sodium sulphate cupper sulphate (10:1) in 5 ml sulphuric acid were found suitable for determination of total Nitrogen, phosphorus and potassium. The steam distillation and the Technicon Auto-Analyzer technique measure similar amounts of ammonium nitrogen. However, the Technicon Auto analyzer technique is easier, rapid, high degree of reproducibility, precise, accurate, reliable and free from human error. The amount of phosphorus measured by the Ascorbic acid/Molybdate method was more accurate than by the Molybdate/ Metavanadate method on Technicon Auto-Analyzer.

The Jean Dumas Method
Jean Dumas (Fig. 1) published his combustion method in Paris back in 1848. This makes the method considerably older than Kjeldahl digestion. While researching the composition of natural oils, Dumas tried combusting his samples, initially to analyse the carbon and hydrogen contained therein. Combustion therefore makes it possible to analyse multiple parameters, while Kjeldahl digestion is only s...
متن کاملCoulometric Determination of Protein Nitrogen. Application to Direct Titration of Kjeldahl Digests.
A coulometric method is described for the determination of ammonia resulting from micro-Kjeldahl digestion of protein samples. The method has been applied to the direct titration of the ammonia in the digests. Routine titration of the ammonia resulting from lOsl. serum samples is described; the method has been applied to 1 -JLI. samples. Results are compared with standard macroand micro-Kjeldah...
متن کاملNitrogen Contents in Food: a Comparison between the Kjeldahl and Hach Methods
The objective of the present work is to compare the nitrogen contents measured by the fast method of Hach and the traditional Kjeldahl method, allowing for the influence of the variation in structure and macronutrient in the food content. Foods with a wide range of protein and moisture contents were selected. The nitrogen content of 25 food samples was measured with both methods. Some studies h...
متن کاملA study of conditions for Kjeldahl determination of nitrogen in proteins; description of methods with mercury as catalyst, and titrimetric and gasometric measurements of the ammonia formed.
The publications on Kjeldahl’s method for determining nitrogen that have appeared since Kjeldahl’s (1) original paper in 1883 perhaps outnumber those on any other analytical method in the same period of time.1 The diversity of papers is attributable to the immense usefulness of the method, to its need for modifications for applications to various types of organic and inorganic compounds, and to...
متن کاملKjeldahl nitrogen analysis as a reference method for protein determination in dairy products.
Measurement of total nitrogen by Kjeldahl analysis is the historical reference method for determination of the protein content of dairy products and is used for both calibration and validation of alternative methods for protein determination. Accurate evaluation of alternative methods is not possible if there is large uncertainty regarding the reference values. When Kjeldahl analysis is used to...
